Pinworm Test-Cellophane (Cellulose Scotch) Tape Preparation Examination

Pinworm Test-Cellophane or Cellulose Scotch Tape Preparation Examination used to identify eggs of adult pinworm at night.

Principle:
The female adult pinworm deposits her eggs during the night on the surface of the skin surrounding the anus. The eggs will adhere to the sticky surface of clear cellophane tape, on which they can be visualized microscopically.

Method:
Place a strip of cellophane tape on a microscope slide, starting 1/2 inch(1.5 cm) from one end and, running toward the same end, continuing around this end across the slide; tear off the strip even with the other end. Place a strip of paper, 1/2 x 1 inch (1.5 x 2.5 cm), between the slide and the tape at the end where the tape is torn flush.
  1. To obtain the sample from the perianal area, peel back the tape by gripping the label, and, with the tape looped adhesive side outward over a wooden tongue depressor held against the slide and extended about 1 inch (2.5 cm) beyond it, press the tape firmly agianst the right and left perianal folds.
  2. Spread the tape back on the slide adhesive side down.
  3. Place name and date on the label.(Note: Do not use Magic transparent tape, but use regular clear cellophane tape)
  4. Lift one side of the tape and apply 1 small drop of toluene or xylen; press the tape down onto the glass slide.
  5. The tape is now cleared; examine under low power and low illumination. The eggs should be visible if present; they are described as football- shaped with one slightly flattened side.
