Hair Perforation Test

A short guide in performing Hair Perforation Test to examine microscopically the presence of conical perforations in infected hairs.

Method:

  1.  Place a filter paper disk into the bottom of a sterile culture dish.
  2.  Cover the surface of the paper with sterile distilled water.
  3. Add a small portion of sterilized hair into the water.
  4. Inoculate a portion of the colony to be studied directly onto the hair.
  5. Incubate at 25 degree Celsius for 10 to 14 days.
  6. Observe hairs regularly by placing them into the drop of water on a microscope slide. Position a coverslip and examine microscopically for the presence of conical perforations of the hair shaft.
