Method:
- Place a filter paper disk into the bottom of a sterile culture dish.
- Cover the surface of the paper with sterile distilled water.
- Add a small portion of sterilized hair into the water.
- Inoculate a portion of the colony to be studied directly onto the hair.
- Incubate at 25 degree Celsius for 10 to 14 days.
- Observe hairs regularly by placing them into the drop of water on a microscope slide. Position a coverslip and examine microscopically for the presence of conical perforations of the hair shaft.
