Principle:
This is a test to detect the presence of Cryptococcus neoformans in a specimen.Negative staining produces a black, semi-opaque background that makes the clear capsule more visible.


Procedure:
  • Mix one drop of specimen (CSF) with one drop of India Ink on a clean slide. 
  • Cover with cover slip. 
  • Examine under LPO and switch to HPO. 
  • If preparation is too dark, it may be diluted with a small drop of H2O.
